anti-human CD31, Clone TR17
Description
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) is an inhibitory coreceptor involved in rebulation of T cell and B cell signaling by a dual immunoreceptor tyrosine-based inhibitory motif (ITIM) that upon associated kinases-mediated phosphorylation provide docking sites for protein-tyrosine phosphatases. CD31 is expressed ubiquitously within the vascular compartment and is located mainly at junctions between adjacent cells. N-terminal Ig-like domain of CD31 is responsible for its homophilic binding, which plays an important role in cell-cell interactions. CD31 is a multifunctional molecule with diverse roles in modulation of integrin-mediated cell adhesion, transendothelial migration, angiogenesis, apoptosis, negative regulation of immunoreceptor signaling, autoimmunity, macrophage phagocytosis, IgE-mediated anaphylaxis and thrombosis. It is one of key regulatory molecules in vascular system.
Properties
The monoclonal antibody ADG5021/L (clone TR17) is a murine monoclonal antibody, subclass IgG1. The antibody has been purified from ascites by protein-A affinity chromatography, Purity > 95% (by SDS-PAGE).
The antibody reacts with CD31 (PECAM-1), a 130-140 kDa type I transmembrane glycoprotein expressed on monocytes, platelets, granulocytes, endothelial cells and stem cells of the myeloid lineage.
Presentation
Vial containing 100 µg /100 µl (ADG5021) or 300 µg /300 µl (ADG5021L) of purified antibody in PBS containing 0.09 % sodium azide (pH 7.2). The IgG concentration is 1 mg/ml. Spin the vial briefly before opening.
Storage and Stability
Store at 4 °C. For longterm storage aliquot and store at -20°. It is recommended to avoid freeze-thaw cycles. The reagent is stable until the expiry date stated on the vial label.

Platelets modulate endothelial cell response to dynamic shear stress through PECAM-1

Meza and colleagues addressed the role of shear stress sensitivity of endothelial cells and platelets. They investigated how platelet-endothelial cell interaction affected endothelial cell inflammatory responses under flow. They found that when endothelial cell CD31/PECAM-1 expression was suppressed by siRNA, platelet adhesion to endothelial cells under low shear stress decreased significantly. They take their findings to suggest that CD31/PECAM-1 mediates platelet adhesion to endothelial cells under shear stress.

Endothelial PECAM-1 and its function in vascular physiology and atherogenic pathology.

Chistiakov and colleagues review the role of CD31/PECAM-1 in endothelial barrier function under physiologic and pathologic conditions with a special emphasis on atherosclerosis. They point out that in vascular inflammation in atherosclerotic lesions, the function of PECAM-1 is impaired, which leads to increased adhesion of neutrophils and other leukocytes to endothelial cells, decreased vascular integrity, and higher leukocyte transmigration to the intima media. Given the intracellular signaling capacity of CD31/PECAM-1 with inhibitory activity, they conclude that in atherosclerosis CD31/PECAM-1-dependent immune suppression is reduced. They conclude that “this in turn facilitates recruitment of leukocytes and supports proatherogenic inflammation.”

Endothelial functions of platelet/endothelial cell adhesion molecule-1 (CD31)

CD31 is also known platelet/endothelial cell adhesion molecule-1 (PECAM-1). CD31 is an inhibitory receptor in circulating platelets and leukocytes, bit is also highly expressed at endothelial cell-cell junctions. Lertkiatmongkol and colleagues review the function of CD31 in endothelial cells, with special emphasis on its role in regulating the vascular permeability. Following inflammatory or thrombotic challenge CD31 functions as an adhesive stress-response protein to both maintain endothelial cell junctional integrity and to accelerate the restoration of the vascular permeability barrier. Certain antibodies directed towards the membrane proximal region of the extracellular domain trigger conformational changes leading to affinity modulation and homophilic adhesion strengthening. Thus “CD31/PECAM-1 might be an attractive target for treating vascular permeability disorders”.
