Highly sensitive chromogenic peptide substrate for plasma kallikrein (PK). Determination of plasma kallikrein activity for research, in-process and quality control.

Biochemical Characteristics:
Vmax = 7.48 µmol/sec KM = 0.175 mM
Assay protocol for the determination of plasma kallikrein
Material required but not provided:
-Dextran sulfate (0.025 g/l)
-Buffer: 50 mM Tris-imidazole buffer, 150 mM NaCl, pH 7.9
Step 1:
Activation of citrated plasma diluted 1:2 with dextran sulfate (0.025 g/l)
=> incubate for 7 min at 0 °C
Step 2:
1.775 ml buffer
0.025 ml Activated plasma or
0.025 ml Enzyme (2.16 units/ml benzoyl-L-arginine-ethylester (BAEE))
=>Determination of OD at 405 nm
Presentation
Amber glass vial containing 25 mg of substrate.
Reconstitution
Reconstitute with filtered deionized water. Shake gently before use.
Storage
May be used by the expiry date given on the label when stored unopened, protected from moisture, in the dark, 2-8°C. Avoid contamination of the reagents by microorganisms.
Shipment of product does not require cooling during the time of transportation.
