murine MAb against human alpha Thrombin
Description
The monoclonal antibody (clone EST4) is directed against human alpha Thombin. Mice were immunized with human vWF purified by gel filtration of plasma cryoprecipitate. Hybridomas were created by fusion of the immunized spleen cells with myeloma cell P3/NS1/Ag4.1.
Properties
The antibody (subclass IgG2a) is directed against epitope in the vicinity of the enzyme active site. It does not bind the thrombin-antithrombin III complex.
Presentation
Screw capped vial containing 0.5 mg of purified antibody in PBS, pH 7.2. The IgG concentration is given on the vial label. Spin the vial briefly before opening. No preservative is added.
Applications
Inhibitory
The antibody inhibits both the clotting and amidase activities of thrombin. Modification of the active site histidine, but not the active site serine, reduced the affinity constant of binding to the antibody.
Storage and Stability
Store the antibody at 2°-8°C. For long-term storage the antibody should be aliquoted and stored at –20°C or colder. It is recommended to avoid freeze-thaw cycles.
